
Epcoritamab Plus R-mini-CVP - Trial Design and Patient Population

Dr. Karr walks host Dr. Kalantri through the clinical problem: patients in their 80s with newly diagnosed DLBCL are often too frail for full-dose R-CHOP or Pola-R-CHP, and historical mini-CHOP data produced modest results — complete response in the low 60s, two-year overall survival rate near 47%. The single-arm trial out of The University of Texas MD Anderson Cancer Center enrolled about 22 patients, median age 80, over a quarter at ECOG performance status of 2, and dropped anthracyclines entirely. Patients received one debulking cycle followed by six cycles of epcoritamab plus R-mini-CVP, testing whether a chemo-light, bispecific-based approach could serve this historically underrepresented population.
